We are leveraging time-based Enterprise Role Request and have put end date as an mandate field. We want to check if there is a way to auto populate end date of the Role request to 31.12.2999.

You can't hardcode end date. You can keep non mandatory on request form which will be considered as no end date.
This is not supported currently and its in Product roadmap
